Brindisi Airport is the most practical air gateway for both Otranto and Santa Maria di Leuca. Bari offers a larger fallback network but adds a longer drive. By rail, reach Lecce on Italy's national network, then use Ferrovie del Sud Est toward Otranto or Gagliano del Capo. Otranto has a regional rail connection; Leuca is reached from Gagliano del Capo by local bus, taxi, or pre-arranged transfer. Drivers arriving through Brindisi or Bari should reserve a vehicle large enough for dive bags and verify one-way or late-arrival fees. International Adriatic ferries into Brindisi can also feed a Salento road trip, but no ferry is required for the destination itself.
A rental car is the most efficient option for a two-base dive trip, especially with wet equipment, dawn check-ins, and inland accommodation. Ferrovie del Sud Est serves Otranto and Gagliano del Capo, with the final roughly 8 km to Leuca handled by bus, taxi, or transfer.
